Is there a trick to loosening the adjustment screw? When I lock the hammer in place to keep it from rotating the adjustment screw doesn't move. Any help would be appreciated.

No trick involved. Is your gun new or used? If used, it could be that the previous owner bottomed it out hard?
Have you tried turning it both ways? If it’s stuck that hard it might require a tear down.

Oh yeah, remember that this gun adjusts in reverse of most other PCP’s. In to reduce, out to increase.
